Hughes was nominated for a Tony Award in 1961 for her work as a supporting actress in The Devil's Advocate.

The actress made her Broadway debut in 1959's The Miracle Worker, understudying the role of Annie Sullivan. Following her Tony Award nomination the next season, she went on to have steady success in Broadway plays for the next two decades.

Her appearances in The Miracle Worker and The Devil's Advocate were followed on Broadway by Dear Me, The Sky is Falling, The Advocate, The Last Analysis, Spofford, The Man in the Glass Booth, Beggar on Horseback, The Prisoner of Second Avenue, Golda, Tribute, The American Clock, Lolita, The Floating Light Bulb and Cafe Crown.
